Constitutional genomic imbalances are known to cause malformations, disabilities, neurodevelopmental delay, and dysmorphia and can lead to dysfunctions in the cell cycle. In extremely rare genetic conditions such as small supernumerary marker chromosomes (sSMC), it is important to understand the cellular consequences of this extra marker, as well the factors that contribute to their maintenance or elimination through successive cell cycles and phenotypic impact. The study of chromosomal mosaicism provides a natural model to characterize the effect of aneuploidy on genome stability and compare cells with the same genetic background and environment exposure, but differing in the presence of sSMC. Here, we report the functional characterization of different cell lines from two familial patients with mosaic sSMC derived from chromosome 12. We performed studies of proliferation dynamics, stability, and variability of these cells using fluorescent in situ hybridization (FISH), sister chromatid exchanges (SCE), and conventional staining. We also quantified the telomere-related genomic instability of sSMC cells using 3D telomeric profile analysis by quantitative-FISH. sSMC cells exhibited differences in the cell cycle dynamics compared to normal cells. First, the sSMC cells exhibited lower proliferation index and higher frequency of SCE than normal cells, associated with a higher level of chromosomal instability. Second, sSMC cells exhibited more telomeric-related genomic instability. Lastly, the differences of sSMC cells distribution among tissues could explain different phenotypic repercussions observed in patients. These results will help in our understanding of the sSMC stability, maintenance during cell cycle, and the cell cycle variables involved in the different phenotypic manifestations.

A doença periodontal é uma afecção comum, relacionada ao aprisionamento de alimentos em diastemas não fisiológicos, em equinos. O tratamento consiste na correção da causa primária, limpeza e desbridamento do sulco gengival, denominado tratamento convencional (TC). Frequentemente antimicrobianos são necessários, pela gravidade ou patogenicidade dos agentes. A terapia fotodinâmica adjuvante (TF) tem sido estudada pelo seu potencial combate bacteriano, sem causar resistência bacteriana. O objetivo deste estudo foi analisar o uso da TF na doença periodontal, experimentalmente induzida, em dentes incisivos de equinos, e compará-la com o TC. O TC não resultou em melhora clínica estatisticamente, tanto em graus como em profundidade, apenas numérica na profundidade aos 30 dias. A TF foi empregada em dentes com profundidade maior da bolsa periodontal que a do grupo TC e, mesmo assim, apresentou melhora clínica já com sete dias, e mais efetiva aos 30, atingindo, em média, o valor considerado normal, três milímetros. A TF apresentou potencial para ser aplicada na rotina, pelo incremento nos resultados, sem causar efeitos colaterais.(AU)


Periodontal disease is a common condition, related to the entrapment of food in non-physiological diastemas in horses. Treatment consists of correction of the primary cause, cleansing and debridement of the gingival sulcus, called Conventional Therapy (CT). Often antimicrobials are requested due to the severity or pathogenicity of the agents. Adjuvant Photodynamic Therapy (PDT), has been studied for its potential bacterial combat, without causing bacterial resistance. The objective of this study was to analyze the use PDT with the experimentally induced periodontal disease in the incisor teeth of horses, and to compare with the CT. The CT did not result in clinical improvement, either in degrees or millimeters. The PDT was used in teeth with a greater depth of the periodontal pocket than the TC group, and even then, showed clinical improvement in only seven days, and more effective at 30, reaching the three millimeter value considered normal on average. The PDT presented the potential to be applied in the routine by the increase in the results without causing side effects.(AU)

Early dietary treatment of phenylketonuria (PKU), an inborn error of phenylalanine (Phe) metabolism, results in normal cognitive development. Although health-related quality of life (HRQoL) of PKU patients has been reported as unaffected in high-income countries, there are scarce data concerning HRQoL and adherence to treatment of PKU children and adolescents from Brazil. The present study compared HRQoL scores in core dimensions of Brazilian early-treated PKU pediatric patients with those of a reference population, and explored possible relationships between adherence to treatment and HRQoL. Early-treated PKU pediatric patient HRQoL was evaluated by self- and parent-proxy reports of the Pediatric Quality of Life Inventory (PedsQL) core scales. Adherence to treatment was evaluated by median Phe levels and percentage of results within the therapeutic target range in two periods. Means for total and core scales scores of PedsQL self- and parent proxy-reports of PKU patients were significantly lower than their respective means for controls. Adequacy of median Phe concentrations and the mean percentage of values in the target range fell substantially from the first year of life to the last year of this study. There was no significant difference in mean total and core scale scores for self- and parent proxy-reports between patients with adequate and those with inadequate median Phe concentrations. The harmful consequences for intellectual capacity caused by poor adherence to dietary treatment could explain the observed decrease in all HRQoL scales, especially in school functioning. Healthcare system and financial difficulties may also have influenced negatively all HRQoL dimensions.

O uso de regimes de corte em estádio vegetativo para os cereais de inverno é interessante quando se busca intensificar a produção de alimento volumoso para ruminantes. No entanto, é necessário se averiguar o impacto desse manejo sobre as características das silagens resultantes. Dessa forma, o objetivo do experimento foi avaliar o valor nutricional e a estabilidade aeróbia de silagens de cereais de inverno, submetidas a regimes de corte no estádio vegetativo, prévio ao corte para ensilagem. Os cereais utilizados foram trigo (Triticum aestivum cv. BRS Gralha Azul), cevada (Hordeum vulgare cv. BRS Brau), aveia-branca (Avena sativa cv. URS Guará), aveia- preta (Avena strigosa cv. Embrapa 139) e triticale (X Triticosecale cv. IPR 11), associados a três regimes de corte: sem cortes prévios (silagem exclusiva); um corte; e com dois cortes em estádio vegetativo e subsequente produção de silagem. Na avaliação nutricional, a cevada apresentou valores baixos de FDA (311,7 e 375,3g kg de MS-1) e altos de NDT (569,2 e 533,4g kg de MS-1) para os sistemas com um e dois cortes, respectivamente. Na avaliação da estabilidade aeróbia, os materiais foram bastante estáveis após a abertura dos silos, com exceção da aveia-branca e da aveia-preta, no regime sem cortes. As silagens de triticale e de cevada apresentaram as maiores estabilidades nutricionais com o aumento no número de cortes.(AU)


The use of cutting regimes in vegetative stage for winter cereals is interesting when it seeks to intensify the forage production for ruminants. However, it is necessary to ascertain the impact of this management on the characteristics of the resulting silage. Thus, the aim of the experiment was to evaluate the nutritional quality and aerobic stability of winter cereal silages submitted to cutting regimes in vegetative stage, before ensiling. The materials used were: wheat (Triticum aestivum cv. BRS Gralha Azul); barley (Hordeum vulgare cv. BRS Brau); white oat (Avena sativa cv. URS Guará); black oat (Avena strigosa cv. Embrapa 139) and triticale (X Triticosecale cv IPR 11); associated with three management strategies: without previous cuts (Only silage), one cut and two cuts in vegetative and posterior silage making. Barley owned low values of ADF (311.7 and 375.3g kg de DM-1) and high TDN (569.2 and 533.4g kg de DM-1) for one and two-cut system, respectively. For aerobic stability, materials were quite stable after opening the silos, with the exception of black and white oat in management without cuts. Triticale and barley silage had the highest nutritional stability with the increase in the number of cuts.(AU)

The aim of this study was to evaluate somatic cell count (SCC), prevalence and etiology of mastitis in a dairy buffalo herd from Analândia, São Paulo State, Brazil, in the dry and rainy seasons. Additionally, antimicrobial susceptibility profile of microorganisms isolated from milk samples was also evaluated. 1,042 milk samples from female Murrah buffaloes in a dairy farm located in Analândia, São Paulo State, Brazil, collected between May 2011 and November 2012 were analyzed. After the mammary gland physical examination, strip cup test and California Mastitis Test (CMT) were performed. Afterwards, 50mL of milk samples from each mammary quarter were collected aseptically for SCC in automatic equipment and microbiological examination. The antimicrobial sensitivity profile to ampicillin, cefoperazone, ceftiofur, enrofloxacin, gentamicin, neomycin, oxacillin, penicillin, and sulfamethoxazole/trimethoprim was evaluated by disk diffusion method. The monthly average temperature and pluviometric index were obtained from "Centro Integrado de Informações Agrometeorológicas" (CIIAGRO) of "Instituto Agronômico de Campinas" (IAC). Milk samples with positive results in the microbiological test showed average SCC of 137,720 cells/mL in the dry period and 190,309 cells/mL in the rainy period. Although a higher number of isolated microorganisms was observed in buffalo milk samples during the rainy period (69/600) compared to the dry period (50/442), the season had no significant effect on the frequency of isolation of microorganisms. The main genera of microorganisms isolated were coagulase-negative Staphylococcus (38.4%), Streptococcus agalactiae (28.8%), and Bacillus spp. (7.56%) during the dry season and Corynebacterium sp. (23.5%), Streptococcus spp. (32.3%), and Streptococcus agalactiae (9.24%) during the rainy period. Multidrug resistance was observed in 30.1% of the isolated microorganisms...


O objetivo do presente estudo foi avaliar a contagem de células somáticas, a prevalência e a etiologia da mastite bubalina nas estações seca e chuvosa em um rebanho de bubalinos do município de Analândia, estado de São Paulo, Brasil. Adicionalmente, verificou-se o perfil de sensibilidade antimicrobiana dos micro-organismos isolados nas amostras de leite das búfalas. Foram avaliadas 1.042 amostras de leite de búfalas da raça Murrah pertencentes a uma propriedade rural localizada no município de Analândia-SP, obtidas no período de maio de 2011 e novembro de 2012. Após o exame físico da glândula mamária, foram realizados o teste da caneca de fundo escuro e o California Mastits Test (CMT); em seguida, foram colhidas, de forma asséptica, amostras de 50mL de leite de cada quarto mamário, para a contagem de células somáticas (CCS) em aparelho automático e exame microbiológico. Também, foi avaliado o perfil de sensibilidade antimicrobiana a ampicilina, cefoperazona, ceftiofur, enrofloxacina, gentamicina, neomicina, oxacilina, penicilina e sulfametoxazol/trimetoprim, pelo método de difusão em disco. A temperatura média e o índice pluviométrico mensais foram obtidos no Centro Integrado de Informações Agrometeorológicas (CIIAGRO) do Instituto Agronômico de Campinas (IAC). Notou-se que as amostras de leite com resultado positivo no exame microbiológico apresentaram CCS média de 137.720 células/mL, no período seco, e 190.309 células/mL, no período chuvoso. Embora tenha se constatado maior índice de isolamentos de micro-organismos nas amostras de leite obtidas no período chuvoso (69/600) do que no período seco (50/442), a frequência de isolamentos não foi influenciada significativamente pela estação do ano...

A Criptococose é uma importante doença infecciosa fúngica, causada por uma levedura do gênero Cryptococcus, que acomete diferentes espécies inclusive o homem. Há poucos relatos na literatura sobre a criptococose pulmonar em cães. O presente trabalho relata um caso de criptococose em um cão apresentando alterações respiratórias, especialmente dispneia. O diagnóstico foi realizado por meio da citologia aspirativa, após toracotomia exploratória, sendo observado um quadro de pleuris grave. O paciente foi tratado durante 90 dias com itraconazol e apresentou, ao final do tratamento, a remissão completa dos sintomas relatados...


Cryptococcosis is a major infectious disease caused by a yeast from the Cryptococcus genre which affects different species including humans. There are few reports related to pulmonary cryptococcosis in dogs. A case of Cryptococcosis in a dog showing dyspnea was described. The diagnosis was done by aspiration cytology after exploratory thoracotomy, and a picture of severe pleurisy was observed. The diagnosis was done by needle aspiration cytology after thoracotomy. Severe pleuris frame was observed. Treatment with itraconazole was performed for 90 days, and at the end of the treatment the dog presented complete remission of reported symptoms...

The identification of pathogens of viral (Rotavirus, Coronavirus), parasitic (Toxocara spp.) and bacterial (Escherichia coli, Salmonella spp., Rhodococcus equi) origin shed in feces, and the virulence profile of R. equi and E. coli isolates were investigated in 200 samples of sand obtained from 40 parks, located in central region of state of Sao Paulo, Brazil, using different diagnostic methods. From 200 samples analyzed, 23 (11.5%) strains of R. equi were isolated. None of the R. equi isolates showed a virulent (vapA gene) or intermediately virulent (vapB gene) profiles. Sixty-three (31.5%) strains of E. coli were identified. The following genes encoding virulence factors were identified in E. coli: eae, bfp, saa, iucD, papGI, sfa and hly. Phylogenetic classification showed that 63 E. coli isolates belonged to groups B1 (52.4%), A (25.4%) and B2 (22.2%). No E. coli serotype O157:H7 was identified. Eggs of Toxocara sp. were found in three parks and genetic material of bovine Coronavirus was identified in one sample of one park. No Salmonella spp. and Rotavirus isolates were identified in the samples of sand. The presence of R. equi, Toxocara sp, bovine Coronavirus and virulent E. coli isolates in the environment of parks indicates that the sanitary conditions of the sand should be improved in order to reduce the risks of fecal transmission of pathogens of zoonotic potential to humans in these places.

RESUMO

Two strains of Nocardia spp. were isolated from bovine milk of two individual bulk tank. Molecular identification classified the strains as Nocardia farcinica and Nocardia cyriacigeorgica. The thermorresistance to boiling of the isolates was carried out and was observed bacterial growth after boiling. Our findings indicate the potential risk of pathogen transmission to humans through contaminated milk with Nocardia spp.

Acylcarnitine profiling by electrospray ionization tandem mass spectrometry (ESI-MS/MS) is a potent tool for the diagnosis and screening of fatty acid oxidation and organic acid disorders. Few studies have analyzed free carnitine and acylcarnitines in dried blood spots (DBS) of umbilical cord blood (CB) and the postnatal changes in the concentrations of these analytes. We have investigated these metabolites in healthy exclusively breastfed neonates and examined possible effects of birth weight and gestational age. DBS of CB were collected from 162 adequate for gestational age neonates. Paired DBS of heel-prick blood were collected 4-8 days after birth from 106 of these neonates, the majority exclusively breastfed. Methanol extracts of DBS with deuterium-labeled internal standards were derivatized before analysis by ESI-MS/MS. Most of the analytes were measured using a full-scan method. The levels of the major long-chain acylcarnitines, palmitoylcarnitine, stearoylcarnitine, and oleoylcarnitine, increased by 27, 12, and 109%, respectively, in the first week of life. Free carnitine and acetylcarnitine had a modest increase: 8 and 11%, respectively. Propionylcarnitine presented a different behavior, decreasing 9% during the period. The correlations between birth weight or gestational age and the concentrations of the analytes in DBS were weak (r £ 0.20) or nonsignificant. Adaptation to breast milk as the sole source of nutrients can explain the increase of these metabolites along the early neonatal period. Acylcarnitine profiling in CB should have a role in the early detection of metabolic disorders in high-risk neonates.

O presente estudo relata a ocorrência de co-infecção entre o vírus da cinomose canina (CDV) e Toxoplama gondii em cães com sinais neurológicos. Amostras de soro e tecido nervoso (pos-mortem) de 21 cães, suspeitos de cinomose canina foram analisadas pela Reação de Imunofluorecência indireta (RIFI) para pesquisa de anticorpos contra T. gondii e N. caninum e por RT-PCR para CDV. Dezessete (80,9 por cento) cães foram positivos para o CDV pela RT-PCR e 8 (38,1 por cento) foram positivos para anticorpos contra T. gondii. Sete cães (41,1 por cento) apresentaram-se positivos para ambos agentes, caracterizando processo de co-infecção. Somente 1 (4,7 por cento) cão foi soropositivo para N. caninum (RIFI=100), entretanto este mesmo animal foi positivo para T. gondii (RIFI=4096) e para CDV (RT-PCR).

Routine diagnosis methods used in bovine mastitis were studied in 55 mares in lactation. The findings of strip cup test, California Mastitis Test-CMT, electronic somatic cell count-CCS, microbiological culture, and in vitro antimicrobial susceptibility profile of isolates were discussed. Streptococcus spp., Staphylococcus spp, and enterobacteria were the most common microorganisms isolated in health and CMT-positive mammary glands. Staphylococcus aureus and Arcanobacterium pyogenes were identified in two mares presenting clinical mastitis. Mean somatic cell count of eight mares without presence of microorganisms in milk was 247.57x10³/mL and 1.621,86x10³/mL in 47 mares with positive microbiological culture. Moderate concordance (63.8 percent) between positive reactions in CMT (1 to 3+) and microbiological culture was observed. Amicacin (78.9 percent), ceftiofur (74.7 percent), sulpha-trimetoprim (69,0 percent) and norfloxacin (69.0 percent), were the most effective drugs, while resistance of isolates was mainly observed against penicillin (64.8 percent), gentamycin (35.2 percent), azithromycin (35.2 percent), enrofloxacin (28.2 percent), and florfenicol (28.2 percent).

An uncommon outbreak of mastitis caused by Arcanobacterium pyogenes in 26 cows is reported. The epidemiological findings, clinical signs, microbiological exams, somatic cell count, in vitro susceptibility profile of strains, efficacy of intramammary treatment and control measures were discussed. Florfenicol (96.2 percent), cefoperazona (92.3 percent), cefaloxin (84.6 percent) and ceftiofur (84.6 percent) were the most effective antimicrobials, and neomicin (27.0 percent) and enrofloxacin (17.4 percent) the least effective antimicrobials.

In the recent years, the wild boar (Sus scrofa) trade has increased in Brazil. The present study aimed to detect Leptospira spp. infection in 308 blood samples from wild boars bred in São Paulo state, Brazil. The microscopic agglutination test (MAT) was performed using 29 serovars. Sixty three (63; 20.45 percent) animals tested positive against the serovars Hardjo (29/63; 46.0 percent), Copenhageni (11/63; 17.4 percent), Pomona (8/63; 12.7 percent), Pyrogenes (4/63; 6.3 percent), Wolfii (4/63; 6.3 percent), Autumnalis (3/63; 4.7 percent), Icterohaemorraghiae (2/63; 3.1 percent), and Hardjo-miniswajezak (1/63; 15.8 percent). These results indicate that captive wild boars were infected with Leptospira spp. and may represent a source of infection to humans and other animals.

The uncommon occurrence of bovine mastitis by Pasteurella multocida is reported in nine cows. Mammary pasteurellosis was predominantly diagnosed predominantly in clinical cases, in dairy cows milked in the presence of calves. Low interference of immunosuppressive or predispose factors and absence of pulmonary signs were observed in animals. In vitro multiple drug resistance of isolates was observed to three or more conventional antimicrobials, especially with use of tetracycline and gentamicin. Epidemiologic findings, clinical signs, and antimicrobial profile were discussed in nine cases of bovine mastitis caused by P. multocida.

The Brazilian Spotted Fever (BSF) is a zoonosis that can be fatal if not trteated. As there are few studies of the BSF in the Paraná State, the occurrence of BSF was serologically investigated in dogs and horses by indirect immunofluorescence assay (IFA), in the North of that State, in animals that are regarded as sentinels for BSF. A total of 241 samples of sera of equine, four of asinine, and 29 of dogs were collected in nine farms of three municipalities in the North of Paraná: Arapongas, Douradina, and Umuarama. From fifteen samples of equine serum with positive titres for RIFI, eight (53.3 percent) had titre of 64 against R. rickettsii, two (13.3 percent) 128 against R. rickettsii, and five (33.3 percent) were reactants for R. parkeri and R. rickettsii, with titres ranging from 64 to 2048, and 128 to 1024, respectively. The results showed that domestic animals, sentinels for BSF, are under low exposition to ticks infected with spotted fever group Rickettsia, indicating low risk of human infection by these agents in the studied area.

RESUMO

Uncommon outbreak of mastitis caused by Pseudomonas aeruginosa strains in 19 cows was reported.Enrofloxacin (73.7%) and danofloxacin were in vitro effective against those strains. In vitro multipledrug resistance of the isolated bacteria was observed to five or more conventional antimicrobials usedfor mammary therapy, including ampicillin (100.0%), cefalexin (100.0%), cloxacillin (100.0%),gentamicin (100.0%), penicillin/novobiocin (100.0%), cefoperazone (52.6%), tetracycline (100.0%),florfenicol (94.7%), and neomycin (73.7%). Epidemiological findings, clinical signs, in vitrosusceptibility profile, control measures, and public health risks were discussed in cows infected by P. aeruginosa.

Susceptibility profile and multiple drug resistance in 158 E. coli strains isolated from urinary tract infection (UTI), pyometra, and feces of dogs were studied. Norfloxacin, ciprofloxacin, and enrofloxacin were the most-effective drugs (>60 percent) for E. coli strains. High rates of resistance to antimicrobials were observed in 60 percent or more of the isolated strains using sulfametoxazole/trimetoprim. Multiple drug resistance for three or more antimicrobials was observed in two (47.1 percent) strains isolated from UTI, seven (13.5 percent) from pyometra, and four (7.3 percent) from feces. From these, 17 (33.3 percent), one (1.9 percent), and three (5.5 percent), respectively, showed multiple resistance to five or more drugs.

O perfil de sensibilidade microbiana e a concentração inibitória mínima (MIC) da azitromicina para 42 cepas de Rhodococcus equi isoladas de potros, no Brasil, e em uma cepa-controle, foi avaliado, respectivamente, pelos métodos de difusão com discos e E-test. A azitromicina apresentou 100 por cento de efetividade in vitro para todas as cepas em ambos os testes. As cepas de R. equi apresentaram MIC90 para azitromicina em valores <1.5µg/ml. Este estudo mostra a alta efetividade da azitromicina em linhagens de R. equi isoladas no Brasil, sugerindo o uso dessa droga como alternativa na terapia da rodococose em potros.
